- Modeling Period4-6 weeks
- Modeling MethodBALB/c mice, weight: 25 ~ 30g, age 6 ~ 8 weeks, erythrhea reflect the sensitively, confirmed by earoscopy no external ear canal and middle ear infection. BALB/c mice were given 1.2 mg of ovalbumin (OVA) in 0.6 ml of phosphate buffer (PBS) and intraperitoneally with 5.14 mg of aluminum hydroxide as an adjuvant for 1 week/2 weeks Sensitization; 2 weeks after the dose of 5 ml / kg body weight by intraperitoneal injection of 20% Uralan anesthesia, aseptic technique 1g / L oVA + PBS mixture 15ul before the tympanic membrane into the tympanum, 24 h after the mouse again Anesthesia, observe the tympanic membrane state, and repeated injection 2 times, ear stimulation; caused by secretory otitis media model. The model animals were subjected to otoscopy every day after injection of OVA solution. The mice were sacrificed 2 days after the second challenge. The thickness of the middle mucosa was measured by HE staining and the eosinophils Cells, plasma cells, toluidine blue staining mast cells.
